Batrachuperus karlschmidti (Amphibian Hynobiidae) Was Found in Qinghai Province

    Abstract:

    This study applied morphological characters and DNA sequence data to identify the two specimens of Batrachuperus collected from Banma County, Golog Tibetan Autonomous Prefecture, Qinghai Province in August, 2012. Two mitochondrial genes Cyt b(754 bp)and COⅠ(582 bp)of the two specimens were sequenced and the sequences of four related Batrachuperus species: B. karlschmidti, B. pinchonii, B. tibetanus and B. londongensis were downloaded from GeneBank. The maximum-parsimony (MP) and maximum-likelihood (ML) phylogenetic trees of these Batrachuperus species were constructed according to Cyt b and COⅠ genes with outgroups Liua tsinpaensis and L. shihi. The phylogenetic trees show that: the two specimens and three known sequences of Batrachuperus karlschmidti form a high supported clade. Based on the phylogenetic trees and the morphological characters that the Body without marble mark, the labial folds well developed and measurements of type, the two specimens were identified as B. karlschmidti, which is found from Qinghai Province for the first tmie.
