    本研究针对内蒙古自治区花鳅属(Cobitis)鱼类多样性及分布现状,以调查过程中采集到的疑似乔氏花鳅(C. choii)为对象,旨在通过形态与分子证据对其进行准确物种鉴定,并据此结合其分布信息,探究其是否属于该区新记录种,进而分析其分布特征与资源状况。2024年7月,在内蒙古自治区呼伦贝尔市辉河、乌尔逊河和巴彦乌拉桥河3条河流分别采集花鳅属标本3尾、4尾和9尾,共16尾。结合15项形态测量指标与基于线粒体COI基因的分子系统学分析,进行物种鉴定。形态指标显示,所有标本均具乔氏花鳅典型特征,包括体侧纵带模糊、L5带(体侧面第五条纵带)由14 ~ 19个斑点组成、雄性胸鳍第二分枝鳍条特化为骨质凸起等。主要形态比例与已报道特征基本一致,但一部分形态学指标变异范围存在一定扩大,如头长/标准长、体高/标准长、尾柄长/标准长、尾柄高/标准长、尾柄高/尾柄长,可能因样本多为未成年个体或地域间的种群形态存在变异。分子系统发育分析结果显示,所有样本与已知乔氏花鳅序列聚为一支,组内遗传距离极低(0.000 0 ~ 0.002 5),支持其为同一物种。综上,本研究通过形态与分子证据,首次确认乔氏花鳅在内蒙古自治区的分布,且其为该地区鱼类分布新记录种。该物种在呼伦贝尔市辉河、乌尔逊河和巴彦乌拉桥河中形成稳定种群,建议加强其栖息地保护与系统监测,以促进区域鱼类多样性研究与保护实践。

    Abstract:

    This study aimed to investigate the diversity and distribution of loaches of the genus Cobitis in the Inner Mongolia Autonomous Region, China. Focusing on specimens tentatively identified as Cobitis choii collected during field surveys, we sought to confirm their taxonomic status through integrated morphological and molecular evidence, determine whether they represent a new regional record, and analyze their distribution characteristics and resource status in the region. In July 2024, 16 Cobitis specimens were collected from three rivers—Huihe River (3 specimens), Ursun River (4 specimens), and Bayan Wula Bridge River (9 specimens)—in Hulun Buir City, Inner Mongolia, China. The species were identified through a combination of morphological examination (15 morphometric characteristics) and molecular phylogenetic analysis based on the mitochondrial COI gene. Morphological measurements followed standards from The Loaches of China. Molecular methods included genomic DNA extraction, PCR amplification and sequencing of the COI gene, sequence alignment by BLAST, and construction of a maximum likelihood phylogenetic tree based on genetic distances to clarify phylogenetic relationships.Morphologically, all specimens exhibited diagnostic characteristics of C. choii, including a faint longitudinal band on the body side, an L5 band composed of 14 ~ 19 distinct spots, and a lamina circularis on the second branched pectoral-fin ray in males (Fig. 1). Key morphological ratios were consistent with previously reported data, though some ratios (e.g., head length/standard length, body depth/standard length) showed wider variation ranges, possibly due to the predominance of juvenile individuals or regional population differentiation (Table 2). Molecular phylogenetic analysis revealed that all obtained COI sequences clustered within a clade of known C. choii sequences, with very low intragroup genetic distances (0.000 0 ~ 0.002 5), strongly supporting their conspecificity (Fig. 2; see detailed genetic distance matrix in Appendix 1). On the basis of congruent morphological and molecular evidence, this study provides the first confirmed record of C. choii in the Inner Mongolia Autonomous Region, which represents a new species record for the area. Stable populations are found across multiple rivers in Hulun Buir City. We recommend enhanced habitat protection and systematic monitoring for this species to support future research and conservation of regional fish biodiversity.
